大孔吸附树脂纯化刺玫果总黄酮工艺研究论文

摘 要

刺玫果学名野蔷薇,又名野刺玫果,山刺玫果,系蔷薇科蔷薇属植物山刺玫的成熟果实。广泛分布于我国的东北地区,尤以黑龙江省贮量最为丰富。现代药理学研究表明:刺玫果含有丰富的营养成分及生物活性物质,具有健脾理气、养血调经、抗衰老、防治心血管疾病、增强免疫及促智等功效。刺玫果中所含的总黄酮类物质具有预防和治疗高血脂、血栓所致的心脑血管疾病的功效,且具有高效、低毒副作用的优势。因此纯化刺玫果中的总黄酮就具有现实的科研意义。

目前,常见的总黄酮纯化方法有水提醇沉法(水醇法)、醇提水沉法(醇水法)、酸碱法、盐析法、离子交换法和结晶法等。实践表明,这些方法也存在一定的局限性。近年出现了一些分离和纯化的新方法,如絮凝沉淀法,大孔树脂吸附法、超滤法、高速离心法等。

本论文采用大孔树脂法纯化刺玫果提取物中的总黄酮。探索洗脱剂、pH值、上样浓度等因素对纯化过程的影响。采用紫外分光光度计法对总黄酮含量进行了分析,最终确定了洗脱剂的浓度75%、上样PH为5,上样浓度为原液浓度,吸附速率为2Bv/h,洗脱速率为1Bv/h。该纯化方法简单,快捷,纯化效率高,进一步拓展了刺玫果的应用领域和范围,为刺玫果的全面和综合利用提供科学的理论依据。

Abstract

Rosa multiflorais is the scientific name of Rosa davurica pall,also known as Wild rosa fruit. Rosa mountain fruit,this fruit is the Rosa mountain fruit of Rosaceae genus Rosa.It is growing in east-north China, especially in province of Heilongjiang. Modern pharmaceutical research shows that Rosa davurica Pall contains rich nutrients and bioactive substances, anti-aging, prevention and treatment of cardiovascular disease, enhances immunity and the efficacy of nootropic, etc. Total flavonoids in the Rosa davurica Pall possess the function of prevention and treatment of high blood lipids, thrombosis dued to cardiovascular and cerebrovascular diseases. It has properties like high-efficiency and the advantages of low toxicity side effects.Thus it is worth in doing research on refining total flavonoids in the fruit.

Recently, there are the several common methods in purifying Flavonoids such as water-extracting alcohol-precipitating,alcohol-extracting water-precipitating,acid-base method, saltdopositing,io-exchange and crystallization,and so on.In practice, there is the limit in all these method reported.Some new methods are investigated like flocculation precipitation, macroporous resin absorption,ultrafiltration,high-speed centrifugea- tion, and so on.

In this thesis, we report extraction of total flavonoids from Rosa davurica pall by using the method of Macroporous resin.It was investigated basing on how the type of Eluant,PH of solution,concentration of samples influences purification.Using Ultraviolet spectrophotometer method to analyze Flavonoids,This method is to know the content of total Flavonoids by measuring Ultraviolet absorption. As a result, it was found that the concentration of Eluant is 75 % ethanol, pH is 5 and the sample concentration is concentration of original solutionl, Adsorption rate is 2Bv / h, Elution rate is 1Bv/h. This method is simple, rapid and high-efficient. This research have obtained some scientific results which will board the application of Rosa davurica pall .
